Development of the Phosphorus Diagnosis Method for P-Fertilization of Cutrus Tree Using 32P Bioassay


Abstract
A ^(32)P bioassay method was applied to develop the phosphorus diagnosis method for P-fertilization of citrus; it is based on the exponential relation between the metabolic uptake of ^(32)P-labelled phosphorus by roots and P-nutritional status in plants. Two year-old Citrus unshiu Marc. cvs. Miyagawa and Okitsu were grown in sand culture or pot culture with different P supply for 6 months, and P-uptake of excised root, P content in leaf and branch growth of tree were measured. There was a correlation between P-uptake by roots excised from 2-year-old citrus and the concentration of phosphorus supplied previously in the sand culture, showing that ^(32)P bioassay can be applied to determine P-nutritional status of citrus tree. By increasing the application levels of P-fertilizer in the pot culture, rates of P uptake by excised root decreased exponentially and the content of available soil P increased. However, P contents in leaves and branch growth of citrus trees were not influenced by P-rates. Therefore, the bioassay assessing the physiological demand for P of plant is more rapid and precise than the chemical analysis and it is feasible to the optimization of P-fertilizer on citrus tree.
